We have examined the effects of activation of protein kinase C (PKC) on the function of human erythrocyte band 3 using the model substrate phosphatidylserine. We report the effects of a novel inhibitor of PKC, diacylglycerol/phorbol ester, on band 3 function in the presence of cyclic nucleotides. Two different functions of band 3 are monitored: (a) an increase in the affinity of band 3 for the anion transport inhibitor, 4,4′-diisothiocyanostilbene-2,2′-disulfonic acid (DIDS), and (b) a decrease in the anion conductance, assayed as the anion permeability of a DIDS-loaded erythrocyte. The PKC inhibitor was found to decrease the affinity of band 3 for DIDS at 0.5 microM DIDS (used in these experiments) in the presence of 4 mM K+ without affecting anion permeability. In contrast, higher (1 mM) concentrations of DIDS caused a decrease in anion permeability without changing the affinity of band 3 for DIDS. At 0.5 microM DIDS, the PKC inhibitor was found to produce an increase in the affinity of band 3 for anion in the presence of 4 mM K+, but no change was observed when this concentration of DIDS was used instead. This DIDS-dependent increase in affinity was concentration-dependent and independent of K+. We propose that the effects of PKC activation on band 3 function may be distinguished by the above results and also by the effects of PKC stimulation on band 3 dephosphorylation by calcium-dependent phospholipase A(2).The 21 patients were selected from the population of patients who died in 2005 at a home health agency in Rochester, Minnesota, and included in this study were those adults who were enrolled at the time of death.
